Processed data for Whole brain cellular activation after mu and NOP receptor agonism identifies differential regional and network consequences

Year: 2026

Abstract: Processed data underlying "Whole brain cellular activation after mu and NOP receptor agonism identifies differential regional and network consequences". TRAP2/Ai9 reporter mice received vehicle, acute morphine (10 mg/kg, i.p.), an escalating twice-daily morphine regimen producing dependence, or the nociceptin/orphanin FQ peptide (NOP) receptor agonist Ro 64-6198 (0.6 mg/kg, i.p.). Neurons active during the labelling window were permanently tagged with tdTomato; whole brains were cleared with MDISCO, imaged intact by light-sheet microscopy, and registered to the Allen Mouse Brain Common Coordinate Framework (CCFv3), yielding cell counts and densities across 198 grey-matter regions (n = 12/12/10/12, 5-6 per sex per condition). This record contains the processed region x animal matrices; the regional statistical results (one-way ANOVA on log10 density with Benjamini-Hochberg correction as the primary analysis, together with sex-stratified, two-way, negative-binomial and globally normalised sensitivity analyses); and the interregional co-activation network results (correlation matrices, differential co-activation tests, global network metrics with a 2-20% density sweep, and cartographic node roles and module assignments). See README.txt for a description of each file. Raw light-sheet imaging volumes are not deposited owing to their size (on the order of terabytes per brain) and are available from the corresponding author on reasonable request.
