Second Harmonic Generation

Year: 2026
Type: standard
Abstract: Second Harmonic Generation (SHG) microscopy is a nonlinear optical imaging technique that produces label-free, highly specific contrast from non-centrosymmetric biological structures—most notably fibrillar collagen. When pulsed laser light interacts with ordered molecular assemblies such as collagen fibrils, photons at exactly half the excitation wavelength are coherently emitted, providing structural specificity without the use of exogenous labels or stains. SHG microscopy enables high-resolution, label-free imaging of extracellular matrix architecture and collagen fiber organization in biological tissues.
