Whole-brain AAV Toolbox BARseq Datasets

Year: 2025
Type: other
Abstract: Our goal is to develop enhancer virus tools that can provide access to select cell classes, subclasses, and types brain-wide. Enhancer-adeno-associated viruses (enhancer AAVs) will be evaluated for specificity and completeness of cell-type labeling using a standardized BARseq spatial transcriptomics approach. Select 20-micron sections from each AAV-injected mouse brain were subjected to in situ sequencing to detect expression of endogenous cell-type marker genes as well as exogenous virally-encoded mRNA (e.g., SYFP). For all datasets, we performed cell segmentation, registration to mouse common coordinate framework, and cell type mapping to the whole mouse brain taxonomy. This data can be used to assess the specificity and completeness of AAV vector expression within cell types and brain regions.
