Comparative In Vitro Effects of Direct Non-Thermal Plasma and Plasma-Activated Medium on U87-MG Glioblastoma and L-929 Fibroblast Cells

Type: preprint
Abstract: Non-thermal plasma (NTP) and plasma-activated medium (PAM) are being investigated as redox-based approaches for glioblastoma. In this manuscript, PAM denotes plasma-treated sterile deionized water subsequently added to complete culture medium. We compared direct NTP exposure and PAM treatment in human U87-MG glioblastoma and murine L-929 fibroblast cells. Direct NTP exposure was applied for 30, 60, or 90 s at a device output setting of 200 V. PAM was tested as undiluted stock (100%) and as 50%, 25%, and 12.5% stock conditions; 10 µL was added to 150 µL of complete medium. MTT-derived metabolic activity was assessed after 72 h. Each condition comprised 24 technical wells within a single plate-based experimental run. Direct NTP exposure for 60 and 90 s reduced MTT absorbance in both cell lines, whereas 30 s produced no clear change. Undiluted PAM stock reduced MTT absorbance in U87-MG cells, whereas the lower PAM stock conditions did not.
