An RNA-seq data resource for time-of-day-dependent cold responses in sorghum and foxtail millet

Year: 2026
Type: article
Abstract: Objectives The purpose of this dataset is to identify transcripts showing time-of-day-dependent responses to low temperature in the panicoid grasses sorghum ( Sorghum bicolor ) and foxtail millet ( Setaria italica ). These data support a larger effort to understand how circadian regulation shapes cold tolerance and its associated fitness costs in C 4 grasses. The strength of this dataset lies in its high temporal resolution sampling under controlled cold treatments initiated at distinct times of day, enabling the detection of rhythmic and cold-responsive transcriptional dynamics. Data description The dataset comprises sequencing data from RNA samples extracted from sorghum and foxtail millet leaf tissue collected after the onset of cold at dawn, dusk, or under control conditions. Plants were grown under a 14-hour light/10-hour dark photoperiod. Samples were collected at selected 4-hour intervals, with each sample consisting of leaf tissue from two plants. Three replicates were collected, with each replicate representing an independent growth trial. poly(A)-enriched RNA was sequenced using the Illumina NovaSeq X Plus platform in a 2 × 150 bp paired-end configuration with a target of approximately 20 million read pairs per sample. FASTQ files are available through the European Nucleotide Archive. Two foxtail millet samples and one sorghum sample were excluded from the data release as a result of poor quality control results after sequencing.
