Role of Superoxide Dismutase 2 (SOD2) in Renal Artery Atherosclerosis: A Clinically Based Perspective
Year: 2025
Type: article
Abstract: Superoxide dismutase 2 (SOD2), a mitochondrial enzyme, plays a critical role in neutralizing reactive oxygen species (ROS), thus maintaining cellular redox homeostasis. Reduced expression or function of SOD2 has been implicated in the pathogenesis of atherosclerosis, primarily due to increased oxidative stress and endothelial dysfunction. In renal arteries, atherosclerosis can lead to renal artery stenosis (RAS), which contributes to ischemic nephropathy, hypertension, and progressive renal dysfunction. This article explores the role of SOD2 in renal artery atherosclerosis, evaluates its potential as a clinical biomarker, and discusses the implications of SOD2-related genetic polymorphisms. We review data from clinical and translational studies that show SOD2 downregulation may enhance vascular calcification and smooth muscle cell proliferation in the renal artery wall. Furthermore, we discuss emerging therapeutic strategies targeting SOD2, including mRNA-based therapies and antioxidant interventions, which may offer renal and vascular protection in high-risk patients.
