Abstract: January image of the NFDI4BIOIMAGE wall calendar 2026, contributed by Sarah Knapp. The original image can be found in BioImage Archive: https://doi.org/10.6019/S-BIAD1673 . Image description: This image highlights PARP15.1, which forms distinct nuclear foci when its ADP-ribosyltransferase domain is active. It shows the reference state of a cell-based assay designed to monitor mono-ADPribosylation (MARylation)-driven foci. Viral macrodomains, such as those from Chikungunya virus and SARS-CoV-2, antagonize this process by reversing host MARylation resulting in spread nuclear GFP signal. Cells were imaged by live-cell confocal microscopy. Channel 1: GFP from mEGFP-PARP15.1. Channel 2: brightfield showing morphology and cell health. Channel 3: nuclear DNA stained with Spy650DNA.
