Comparative analysis of seven Buxus (Buxaceae) species provides insight into plastid genome evolution, ndh pseudogenization, and inverted repeat dynamics

Type: dataset
Abstract: Buxus (Buxaceae) comprises 101 species, but plastome resources for South Asian taxa were unavailable. We de novo assembled five Buxus plastomes—B. papillosa, B. wallichiana, B. harlandii, B. sempervirens, and B. austroyunnanensis—and compared them with two published genomes (B. microphylla and B. sinica var. parvifolia), providing first plastome resources for B. papillosa and B. wallichiana. Plastomes had typical quadripartite structure (150,915–161,655 bp). Inverted-repeat dynamics revealed four genome types, with B. sempervirens most contracted and B. wallichiana most expanded. SSRs were AT-rich mononucleotide repeats mainly in noncoding regions; codon usage and amino acid frequencies were conserved. Four intergenic spacers (rps16–trnQ-UUG, ycf4–cemA, ccsA–ndhD, trnE-UUC–trnT-GGU) were highly variable for DNA barcoding. Phylogenetic analyses supported B. papillosa + B. wallichiana and a four-species group, while B. austroyunnanensis placement remained unresolved. ndhF loss was shared by sister species; additional ndhD/G/H/I pseudogenization occurred only in B. wallichiana. Data provide genomic foundation for Buxus evolution, diversification, and conservation.
