Keyora Skin Systems Nutrition - Collagen Tripeptides EP-11: The Five-Step Collagen Trust Algorithm: Four-Object Molecular Quantification, Analytical Verification, and Preparation-Matched Human Evidence
Year: 2026
Type: article
Abstract: Background Collagen hydrolysate labels often emphasize total milligrams, although equal ingredient masses may conceal different peptide populations. Peptide-class fractions, amino acid composition, and intact peptide concentrations represent analytically distinct quantities; none independently demonstrates clinical efficacy. Objective and Scientific Framework This narrative synthesis introduces the Keyora Four-Object Collagen Composition Matrix and operationalizes it through the Keyora Collagen Tripeptide Composition and Trust Algorithm: IDENTIFY, QUANTIFY, VERIFY, MATCH, and DECIDE. Five chapters integrate material identity, tripeptide enrichment, amino acid and sequence chemistry, human evidence, and reproducible selection criteria. Mechanistic and Analytical Foundations Native collagen contains Gly-X-Y triple-helical domains; denaturation and enzymatic hydrolysis generate heterogeneous peptide populations. Keyora declares 1,500 mg of tilapia-scale hydrolysed collagen powder per two-tablet serving, a 30% total tripeptide fraction, combined Gly + Pro + Hyp above 50%, and supplier-reported mean molecular mass near 459 Da. A verified 30% ingredient mass fraction would represent approximately 450 mg of total tripeptides, not 450 mg of intact Gly-Pro-Hyp (GPH). The GPH concentration remains independently unquantified. Amino acid analysis determines residue abundance, whereas calibrated, sequence-selective LC-MS/MS, authentic reference standards, and batch-linked analytical records establish intact peptide identity and quantities. Experimental and Human Evidence Yamamoto (2016) compared porcine collagen preparations with reported tripeptide peak-area fractions of 91.80%, 50.66%, and 1.59%, and approximately 34%, 7%, and 0% GPH, respectively. Four men received 80 mg/kg of each preparation; tripeptide-rich products produced higher circulating GPH and Pro-Hyp exposure, with plasma GPH peaks approximately 30-60 minutes after ingestion. Cultured fibroblast studies of Pro-Hyp implicate STAT3 phosphorylation, HAS2 expression, and hyaluronan synthesis, providing mechanistic plausibility rather than clinical validation. Kim (2018) randomized 64 women to 1,000 mg/day of a sutchi-catfish-skin collagen preparation containing over 15% tripeptides and approximately 3% GPH, or placebo, for twelve weeks. Hydration improved versus placebo at weeks six (p < 0.001) and twelve (p = 0.003); elasticity R2 (p = 0.025), R5 (p = 0.027), and selected wrinkle measurements improved at twelve weeks. These results pertain to the tested preparation. Integrated Scientific Interpretation The algorithm differentiates declared composition from verified mass fractions, sequence-specific concentrations, and human exposure. Its evidence-matching step evaluates species, tissue source, peptide profiles, dose, participants, comparator, study duration, and measured endpoint before classifying direct, composition-comparable, or general evidence. Evidence Boundaries and Conclusions A 2025 synthesis of 23 trials (1,474 participants) found favorable pooled skin outcomes, but significant benefits did not persist in higher-quality or non-industry-funded subgroups. A 2026 synthesis of 35 trials (2,534 participants) reported hydration improvement (SMD 0.44) and reduced transepidermal water loss (SMD -0.39); elasticity improved after influential-study exclusion, while pooled wrinkle findings lost significance after outlier exclusion. Neither pharmacokinetic findings nor external trials establish efficacy for the finished Keyora preparation. The five-step algorithm offers a transparent, auditable pathway from molecular characterization to appropriately bounded evidence-informed collagen comparisons.
