Abstract: L-carnitine plays an important role in the mitocondrial uptake of long chain fatty acids in mammals. It has recently been shown thatthis compound has a marked cholesterol-lowering effect when it is used in conjunction with lipid rich diet. The pharmacological effects of supplementation of [80 mg/(kg ü d)] on established hypercholesterolemia and atherosclerotic lesions were studied in cholesterol-fed rabbits. The administration of this atherogenic diet during 126 d provoked high level of blood cholesterol [(18,43 Á 6,77) g/L, n = 6] and ath~romatosis (50,25%) on the aorta intima layer. L-carnitine supplementation together with this diet decreased the blood cholesterol [(10,39 Á 2,07) g/L, n = 6] and the aorta atherosclerotic lesions (44,25%). After hypercholesterolemia and aortic dangerwere established, the atherogenic diet was substituted for standard diet and the L-carnitine effect on the regression of these events was studied during other 65 d. The L-carnitine supplementation in this stage produced remarkable increase in triglycerides from VLDL+LDL modified fraction and esterification process of cholesterol from HDL; at the same time changes in fatty acids composition esterified to cholesterol and significant regression atheromatous plaque were observed. These results show a L-carnitine positive effect on hypercholesterolemia reduction and aortic fatty streaks.
