Abstract: This chapter focuses on the synthesis and biological properties of the relatively small family of pyrido[4,3-b]carbazole alkaloids, exemplified by ellipticine, 9-methoxyellipticine, 9-hydroxyellipticine, and olivacine, and of the much larger number of structural analogs that have been synthesized and studied following the initial discovery of the antitumor properties of these alkaloids. Ellipticine and 9-methoxyellipticine have been isolated from Ochrosia acurninata stems and from in vitro callus cultures derived from the stems of Ochrosia elliptica. The diversity of structural variation and substitution pattern in these molecules continues to fascinate and challenge synthetic chemists, while the myriad of pharmacologic activities have been revealed by the ellipticines to maintain their attraction to biochemists and pharmacologists.
