Development of safe harbor CRISPR inhibition and activation tools for systematic biological inquiry in zebrafish

Description: The shared biology between human and zebrafish has facilitated the establishment of this fish as a powerful model system in which to functional explore the function of genes implicated in disease and the sequences that control their timing and location of activities. The development of new CRISPR genome engineering technologies allow the upward and downward tuning of gene activities in mammalian systems but have not yet been fully implemented in zebrafish. We will develop a new system for zebrafish in which this kind of gene function modulation is rendered systematic and straightforward and we support our proposal with key proof of principle data.
