Abstract: A reduction in monomer consumption in the solid phase synthesis of oligodeoxyribonucleotides has been achieved by introducing a repeat coupling approach. Additionally, a method of anion exchange HPLC- purification of the synthesized nucleotides at room temperature has been developed in which a highly volatile buffer was used as the eluent. An alternative approach of HPLC purification of oligonucleotides is also reported.
