Abstract: The objective of this research was to demonstrate the applicability of PLGA nano -spheres for transdermal drug deliveries.By applying spherical crystallization with emulsion solvent diffusion method in an aqueous PVA solution, the pro -vitamin C loaded and the coumarin -6 loaded PLGA nano -spheres with an average particle size of 215 nm were prepared and tested via the human skin biopsies in a modified Bronaugh diffusion chamber. The coumarin -6 loaded PLGA nano -spheres showed excellent capability of passing through skin pores and had a superior cutaneous penetrability to deep -epidermis, both in speed and quantity compared to the coumarin -6 dispersion liquid at a much higher concentration. The pro -vitamin C loaded PLGA nano -spheres were also found to be much more effective in supplying reduced vitamin C to deep-skin than provitamin C emulsion. In addition, the PLGA nanospheres could prolong the retention of reduced vitamin C for better skin care results.Furthermore, inhibitory effects of pro-vitamin C loaded PLGA nano-spheres on DNA double-strand breaks by UV-A and UV-B irradiations were confirmed. The PLGA nano-sphere was proven to be a viable vehicle for transdermal drug delivery application.
