Quantitation of low abundance cytokines in chronic autoimmune diseases using SMC® high sensitivity immunoassays 3112

Abstract: Description Quantitation of biomarkers in blood is crucial for the understanding of autoimmune diseases caused by chronic inflammatory response. Pro-inflammatory as well as anti-inflammatory cytokines have been implicated in the pathogenesis of a variety of diseases. In basic research, standard immunoassays, such as an enzyme-linked immunosorbent assay (ELISA), have provided valuable understanding of both normal and pathological processes for many decades. However, in more recent decades, new immunoassay technologies have uncovered numerous cytokines in blood that were once undetectable using traditional ELISAs. In this study, we have developed ultrasensitive immunoassays, using SMC® technology, to meet this new challenge of quantifying low abundant proteins in blood. Furthermore, we have developed the next generation FemtoQuest™ system, which allows detection of two analytes in a single assay well, thus saving precious samples. The SMC® 2-plex IL-17A,IL-17F; IL-1α,IL-1β; IL-22,IL-23; and TNFα,IL-6 assays consistently demonstrated LLoQ as low as 0.07 pg/mL. The dynamic range of these ultrasensitive SMC® kits allowed quantitation of both healthy and disease blood samples in a single assay. We report that an elevated level of several analytes were observed in rheumatoid arthritis, systemic lupus erythematosus, or Sjögren’s syndrome serum or plasma as compared to healthy samples. In summary, SMC® high sensitivity immunoassay kits provide a powerful research tool for autoimmune research. Topic Categories Basic Autoimmunity (BA)
