The Identification of Ozonolysis Products from Bisphenol Analogues and their Binding Affinity to Estrogen-Related Receptor Gamma

Year: 2026
Type: dissertation
Abstract: Current chemical production levels are causing elevated chemical exposure in humans, both from the produced compounds and their transformation products. However, understanding the health risk from transformation products is difficult due to an absence of standards for chemical and biological testing. Here, I established a novel framework to assess the toxicity of transformation product mixtures by combining ozonolysis and protein-guided nontargeted analysis. Bisphenol A alongside five of its analogues were selected to develop and validate the framework. Dozens of ozonolysis products were formed from the bisphenols including previously reported and newly found compounds. Aside from some weakly binding bisphenol AF products, none of the ozonolysis products bound to estrogen-related receptor gamma protein, demonstrating the general detoxification of bisphenols through ozonolysis. This research validated the framework, identified novel transformation products of bisphenols through nontargeted LC-HRMS, and discovered the decreased protein binding ability of transformation products compared to parent bisphenols
