NPM1-mutated AML: FLT3-ITD vs FLT3-TKD by DNMT3A/TET2 status - multi-cohort public-data analysis (code and results)

Abstract: Reproducible code, pre-specified protocol, data provenance and reference results for a multi-cohort public-data analysis (TCGA-LAML and Beat AML via cBioPortal; AMLSG) testing whether DNMT3A/TET2 co-mutation modifies the overall-survival difference between FLT3-ITD and FLT3-TKD in adults with NPM1-mutated acute myeloid leukemia. A single command downloads the public source data, harmonises variants, runs cohort-stratified Cox models with sensitivity analyses and a 2,000-replicate bootstrap, regenerates all tables and figures, and checks the re-run against the reported estimates.
